Waters has found that for quantitation of phospholipids, Oasis PRiME HLB works well if following a method that was first published using a Waters Ostro plate:

Extraction of Phospholipids from Plasma using Ostro Sample Preparation

This method can be scaled up to larger Oasis PRiME HLB sorbent bed masses.

Alternatively, if quantitation is not required, the following steps can be evaluated:

1. Pretreat plasma samples 1:1 with a solution of 4% phosphoric acid in water.

2. Load pretreated samples.

3. Wash with 5% methanol in water.

4. Elute 1: 100% acetonitrile (to elute unwanted hydrophobic compounds)

5. Elute 2: 100% methanol

Note: If all phospholipids do not elute in 100% methanol in the elute 2 step, you can add some water to the elute 2 step, such as 90:10 methanol/water to try to get more phospholipids to elute.
