Do I need to perform protein precipitation on my plasma samples before I load them onto Oasis PRiME HLB cartridges or 96-well plates? - WKB227466

Article number: 227466

ENVIRONMENT

  • Sample pretreatment
  • Hydrophilic-lipophilic balance (HLB)
  • Oasis PRiME HLB cartridge
  • 96-well plate

ANSWER

No

ADDITIONAL INFORMATION

Proteins are expected to pass through Oasis PRiME HLB packing material because they cannot enter the pores of the packing material (they are excluded).

Waters recommends that you dilute plasma samples 1:1 when using a diluent of 4% phosphoric acid in water. This is designed to disrupt protein binding. High pH may also be used to disrupt protein binding prior to loading. In some cases, if low pH or high pH sample pretreatment does not fully disrupt protein binding, protein precipitation (PPT) may be performed prior to loading. In most cases, PPT is not required.

Also see the Oasis Reference Cards for method information, sample pretreatment guidance, and more.
