A protein trimer may be similar to the SARS-CoV-2 spike protein, which is a ~540 kDa homotrimeric glycoprotein.

Waters has done some studies on the spike protein using a reducing protocol (see the following links) and identified 42 major glycan peaks.

In the reducing protocol, we use DTT to help digest the molecules with multiple disulfide bonds.

The protocol is optimized for glycoprotein with a concentration of 1.5 mg/mL.

*You can use the reducing protocol as a starting point and monitor the intact mass to determine if the deglycosylation is complete.
