ANSWER

When using a 96-well positive pressure manifold, one must use the square well collection plate (WAT058957 or 186002482) below the sirocco plate (186003873 or 186002448) and not the round well collection plate. Otherwise, the valve mat forms a seal in the round wells in the collection plate and the sample will not process properly.  

There is no collection plate or seal/cap for the collection plate included with the Sirocco plate. The cap mat included in the "kit" is to cover the Sirocco plate for the mixing and vacuum/centrifugation steps.
