Traditionally, Waters PIC reagents are designed to be used in isocratic methods only, because the PIC reagent needs to adsorb to the hydrophobic packing. 

If a gradient is used, the increased organic washes off some of the PIC reagent, leading to potential reproducibility issues with retention, selectivity, or both.

You may experiment using Waters PIC regents in a gradient method, with the understanding that these were not designed for use in gradient methods.
