What is the recommended cleaning procedure for a Spherisorb SAX column? - WKB70452

Article number: 70452

ENVIRONMENT

  • Column care and cleaning
  • Spherisorb SAX

ANSWER

The recommended cleaning procedure is as follows: Flush with 10 column volumes of slightly basic (~pH 7.0) buffer. To prepare to flush with pH 7 wash, flush the lines without the column inlet connected or divert the flow to waste for about 30 minutes prior to hooking up the column for the cleaning.

The column can also be cleaned with 100% organic, but make sure to remove all salt buffers from the system prior to flushing in any organic to avoid precipitating buffer in the lines and column.

Additional cleaning procedure: Rinse with 20 column volumes buffer (pH 3.0), 20 column volumes water, 20 column volumes methanol, 20 column volumes methylene chloride, 20 column volumes methanol, 20 column volumes water, and then 20 column volumes buffer.

ADDITIONAL INFORMATION

Observe any solubility concerns the sample may exhibit and do not use a cleaning solution that would precipitate the sample residues on column, out of solution.
