ANSWER
When a new CORTECS HILIC column is used, it must be flushed for 50 column volumes with 50:50 acetonitrile/water and 10 mM of the buffer used in the method.
After the 50:50 acetonitrile/water with 10 mM buffer step, equilibrate the column for 20 column volumes in the initial conditions of a gradient or the isocratic mobile phase.
