PROCEDURE

  1. Go to the Waters On-line Preparative Columns Calculator.
  2. Choose any of the following buttons on the lower portion of this webpage:
    • Analytical to Prep Gradient Calculator:
      • Choose the "Gradient Calculator" tab to scale:
        1. Gradient
          • Specify the details of the analytical method on the left-hand side.
          • Specify the proposed prep column dimension on the right-hand side to scale a gradient.
        2. Flow Rate
        3. Injection Volume
        4. Mass Load (per injection)
      • Choose the "Method Optimization" tab to calculate:
        1. Pressure values for different solvents
        2. Estimated column back pressure
        3. Various other calculations
      • Choose the "Dwell Volume" tab:
        1. This webpage shows how to calculate system dwell volume.
    • Tubing Back Pressure and Splitter Calculator:
      • Choose the "Tubing Backpressure" tab to calculate tubing pressures.
      • Choose the "Tubing Splitter Calculations" tab to calculate various splitter values.
    • Focused Gradient Calculator:
      • Choose the "UPLC to Prep focus gradient" tab to calculate:
        1. Optimized gradient focus to selectively elute target analyte with a slower gradient change in the area where the target peak needs the most resolution.
        2. If needed, specify a new flow rate for the focus gradient using the field in the "Override Focus Flow Rate" box.
        3. Select Language using the "Please Select Language" button on this webpage:
