Why do I see a negative peak at the total inclusion volume when running my ACQUITY UPLC BEH SEC column with UV detection and acetonitrile in the mobile phase? - WKB229118

Article number: 229118

ENVIRONMENT

  • UltraPerformance Liquid Chromatography (UPLC)
  • Bridged ethylene hybrid (BEH)
  • Size exclusion chromatography (SEC)
  • Protein separation
  • Negative peak
  • Inclusion volume
  • ACQUITY UPLC BEH SEC

ANSWER

A negative peak may be seen at the total inclusion volume if the sample diluent has less UV absorbance than the mobile phase.

ADDITIONAL INFORMATION

The total column inclusion volume is the combined value of the pore volume and the interstitial volume of the column.
