ANSWER

Run the column at 0.1 mL/min for 50 minutes in 50:50 acetonitrile/water. This can be flowed to waste.

While the column is in the 50:50 acetonitrile/water, allow the column temperature to reach 55 °C.

After 50 miniutes, flush the column with the starting conditions and slowly raise the flow rate to 0.5 mL/min.  

To "break in" a reverse-phase column, some derivatization blanks should be injected while running the standard 4-minute Kairos gradient. Inject 2x's derivatization blanks, and then follow with two to four injections of the derivatized amino acid standard.

ADDITIONAL INFORMATION

It is normal for reverse-phase columns to need four to six injections of standard to condition the new column to run the method.
