ANSWER
Before using it, you need to check the following.
- Column end fitting specification
- Available mobile phases and additives, mobile phase pH, salt concentration, column temperature, maximum pressure
- Column cleaning method
- External dimensions of the column
Column end fitting specification
Waters has adopted two types of specification.
- Waters type—ferrule tip tube length of 3.30 mm
- Parker type 2.28 mm
Waters UPLC columns are the Parker type. The end-fitting specification may differ depending on the manufacturer, series, and size of the column, so make sure to check before use.
If the fittings used are not appropriate, problems such as peak broadening and poor peak shape due to dead volume may occur. Also, leakage from the ferrule parts may occur.
Available mobile phases and additives, mobile phase pH, salt concentration, column temperature, maximun pressure
The mobile phase and instrument conditions that can be used may differ depending on the column manufacturer and series.
When using the functional groups of the same solid phase, refer to the instructions published by the manufacturer.
Also, check the mobile phase conditions recommended for the HPLC or UPLC system used.
Column cleaning method
Check the cleaning method, as well as the mobile phase conditions above.
External dimensions of the column
Ensure that the column size fits correctly in your column heater or column manager.
Depending on the external dimensions of the column, these instruments may not be able to accommodate it, and the column may not be able to be heated or cooled properly.
