ANSWER

The application notes use a standard of the AAV serotype of interest to calibrate the detector response and the Waters BEH450 SEC protein standard mix with L-tryptophan spiked into it to make sure the column is separating a protein mix of known molecular weights.

ADDITIONAL INFORMATION

The separation of the AAV from large multimer aggregates and lower molecular weight fragments is to determine the percentage of AAV monomer vs. smaller fragments.
