Problem getting mAb charged variant standard retention on BioResolve SCX mAb column - WKB195920

SYMPTOMS

• No retention of mAb charged variant standard on the BioResolve SCX mAb column

ENVIRONMENT

• BioResolve SCX mAb
• Charged variant standard
• Column care and use

CAUSE

The mAb charged variant standard is not optimized for benchmarking with the IonHance CX-MS pH concentrate and gradients. The eluent strength and conditions are not optimized for the mAb charged variant standard.

FIX or WORKAROUND

1. The mAb charged variant standard should be run with the BioResolve CX pH concentrates and the benchmarking gradients used with the BioResolve CX pH concentrates.
2. If using the IonHance CX-MS pH concentrates with the BioResolve SCX mAb column, the column should be benchmarked with the Humanized NIST mAb standard (186009125).
