If the method is a gradient method and none of the analytes elute at the initial mobile phase conditions, this may be possible. The main thing to consider in this case is mass overload. If too much mass is loaded in the 500-µL injection, you would expect a "shark fin" peak shape because analyte will elute earlier and earlier at the front side of the peak the higher the on-column mass is.
