How can I get a UV signal at 260 and 280 nm when my AAV concentration is 10^12 to 10^13 Vg/mL? - WKB124738

Article number: 124738

ENVIRONMENT

  • Size exclusion chromatography (SEC)
  • AAV
  • UV
  • RI

ANSWER

In SEC separations, injecting a larger sample volume will provide some increase in sensitivity, although at some point resolution will be impacted.

ADDITIONAL INFORMATION

Decreasing the flow rate can also improve sensitivity a bit. UV absorbance peak sensitivity will ultimately be a function of the detector characteristics and the abundance of aggregates and fragments in the samples being analyzed. RI will generally not increase sensitivity versus UV absorbance.
