What are the recommended initial column conditioning steps when performing insulin testing with Insulin HMWP SEC, 125Å, 10 µm, 7.8 mm X 300 mm, 5-150K column (WAT201549)? - WKB121979

Article number: 121979

ENVIRONMENT

  • Insulin testing
  • High-molecular-weight protein (HMWP)
  • Size exclusion chromatography (SEC)
  • Column conditioning

ANSWER

1. Purge the column with 5 column volumes (50 mL) of HPLC-grade water.

2. Purge the column with 5 to 10 column volumes of the mobile phase used for the your insulin testing method until a stable baseline is observed.

ADDITIONAL INFORMATION

If the same column is used for more than one application, it is recommended that you make a 100-µL injection of a sample that is used for the new method, in order to clear the injector of any residual solvents or samples from a previous method run on the same column.
