Why do I see a large glutamine peak in my AccQ•Tag chromatography when I use GlutaMax in my cell culture media? - WKB115261

ENVIRONMENT
- AccQ•Tag
- GlutaMax
- Glutamine
- Cell culture
- Chromatography
- Derivatization
ANSWER
GlutaMax cell media supplement is a high source of amines—a dipeptide. When the cell culture media sample is derivatized with AccQ•Tag reagent, the amines are derivatized.
ADDITIONAL INFORMATION
The cells digest the GlutaMax cell media supplement slowly by producing peptidases that break the dipeptide down into L-glutamine and L-alanine. The additional high amounts of L-glutamine can be detected because they derivatize and the excess glutamine elutes in the same place as native glutamine.
